چکیده

The current ISO standard method for detection of Enterobacteriaceae (21528-1:2004) includes enrichment in EE broth which has been shown to be inhibitory to some members of this family, notably Cronobacter spp. A shortened procedure omitting the EE broth has been proposed, however competition from Gram-positive flora may be detrimental to the effective recovery of low levels of target organisms in some sample matrices. In this study we investigated novel cost effective modifications, designed to improve ISO 21528-1:2004 for the detection of Enterobacteriaceae. Initial experiments used a worse-case scenario involving stressed Enterobacteriaceae strains known to grow poorly in laboratory media as well as representative background competitors from powdered milk. The interaction between the Enterobacteriaceae and their competitors was characterised and additives to enhance the growth of target strains over non-target strains were investigated. Supplementation of BPW with 40 microM 8-hydroxyquinoline, 0.5 gL(-1) ammonium iron(III) citrate, 0.1 gL(-1) sodium deoxycholate and 0.1 gL(-1) sodium pyruvate (BPW-S) improved the recovery of Enterobacteriaceae from artificially and naturally contaminated samples. This improvement of the pre-enrichment broth may also be of interest for methods designed to detect specific foodborne pathogens belonging to the Enterobacteriaceae (e.g. Salmonella spp., Cronobacter spp.) that require a pre-enrichment step in BPW.

Rapid detection of sublethally impaired cells of Enterobacteriaceae in dried foods.

Samples of dried foods, feeds, and drugs were examined for Enterobacteriaceae by using buffered glucose-Brilliant Green-bile broth as the final enrichment medium and deep tubes of violet-red-bile-glucose-agar for confirmation. The pre-enrichment treatments used for the resuscitation of sublethally impaired cells were overnight incubation in lactose broth versus incubation for 1 to 6 hr at room ...

Multiplex detection of nine food-borne pathogens by mPCR and capillary electrophoresis after using a universal pre-enrichment medium

Routine microbiological quality analyses in food samples require, in some cases, an initial incubation in pre-enrichment medium. This is necessary in order to ensure that small amounts of pathogenic strains are going to be detected. In this work, a universal pre-enrichment medium has been developed for the simultaneous growth of Bacillus cereus, Campylobacter jejuni, Clostridium perfringens, Cr...

Evaluation of a new enrichment broth for detection of Cronobacter spp. in powdered infant formula.

The aim of this study was to investigate the potential of using Al-Holy-Rasco (AR) medium, a novel broth for detection and isolation of Cronobacter spp. in infant formula milk (IFM). The new medium's composition is generic brain heart infusion broth with the addition of 1% NaCl, 15% sucrose, and 0.80 g/liter sodium deoxycholate as selective ingredients. AR broth outperformed Enterobacteriaceae ...

Rapid quantitative detection of Listeria monocytogenes in chicken using direct and combined enrichment/qPCR method

Listeria monocytogenes is a species of foodborne pathogen often related to foods, such as poultry, ready-to-eat products, fruits, and vegetables. The culture method is a standard procedure for the detection of bacteria in food products. The real-time quantitative PCR (qPCR) technique can be used for the quantification of foodborne pathogens. The current research was aimed to assess...

Rapid detection of Salmonella in foods using a combination of SPRINTTM, MSRVTM and Salmonella Latex TestTM

A new procedure for rapid detection of Salmonella in foods, based on the combination of SPRINTTM, MSRVTM and Salmonella Latex TestTM, was evaluated. SPRINTTM is a system to reduce the preenrichment and selective enrichment steps to 24 hours. MSRVTM is a semi-solid selective media for detection of motile Salmonella. Salmonella Latex TestTM is a rapid latex agglutination test for Salmonella. Usin...
